Excellent campaign concept! I'll add it to the "sounds like fun to run" stack, and maybe some day my group will bite.
I'd like to add The Savage World of Solomon Kane to the list of potential RPGs. The technology level is the same or at least close enough. You'd just need to move the setting.

But seriously, I do wish I'd thought of this myself. It's positively brilliant.
Oh, and I thought I'd add that Melissa de la Cruz's Blue Bloods novels have a very interesting take on the Croatan story (and vampires in general). There was an Adrian Paul movie on Sci-Fi recently called "Wraiths of Roanoke" (I've since seen it in Blockbuster under another title). I haven't seen it, but it might be good source material.

And while on the subject of inspirational movies, Ginger Snaps is a lovely little thing with werewolves on the frontier.
And M. Night Shamalan's "The Village", can be used for mood. Isolated town, local superstitions, etc.
And there was a thing a while back with an evil tree, but I've lost the name.

Awesome concept, though the setting described in the article needs some historic tweaking. Sleepy Hollow is immediately POST-Revolutionary US. New York hadn't been New Amsterdam for quite some time.
